Block port in use

After I find a specific port with a netstat, can I block or disconnect it, even while it's in use?

I tried on skype conversation with Jokke's firewall, but it's not supporting ports already in use.

Is there any other method ? ;)

+ I want to add this to my script, so it can't be done with other program, eg. Emsa port blocker.

Why do you want to do that?

Why not using a proxy, firewall?

I'm using push-to-talk software, which doesn't allow to switch server, before you disconnect from last and wait 30 sec. But when I disconnect using port block, it allows me to do that instantly. I tried with IPseccmd and I know it works, but I was wondering if there's a similar method in AutoIt.
