Ok this is what I have:

______________________        ________
|Readonly Input box___|       |Button A|

I have the input box as

GuiCtrlCreateInput("Readonly Input box", 1, 1, 70, 20, $es_readonly)

And i want it to just be normal after the button is clicked.

You maybe want try activate/deactive way...

#include <GUIConstants.au3>

GuiCreate("Test", 244, 76)

$btnState= $GUI_DISABLE

$Input = GuiCtrlCreateInput("text", 10, 20, 130, 30)
GUICtrlSetState(-1, $btnState)

$Button = GuiCtrlCreateButton("Enable", 150, 20, 60, 30)

While 1
    $msg = GuiGetMsg()
    Case $msg = $GUI_EVENT_CLOSE
    Case $msg = $Button
      If $btnState = $GUI_DISABLE Then
         GUICtrlSetState( $Input, $GUI_ENABLE)
         GUICtrlSetData( $Button, "Disable")
         $btnState = $GUI_ENABLE
         GUICtrlSetState( $Input, $GUI_DISABLE)
         GUICtrlSetData( $Button, "Enable")
         $btnState = $GUI_DISABLE
