using transparent png with imagesearch?

I need help , i have some transparent png and some do work but only if there like really small , but my 44x27 transparent png seems imagesearch cant find it for some reason even if i mess around with the tolerance level? is there something im missing

does bit-depth have anything to do with it i noticed the one that works is 24bit depth and the one that does not is 32bit-depth, if that is indeed the case how would i convert it to 24bit depth with photoshop
