pixel detection help/gui making.

i wanna "make/have made" a fishing macro for "Trove" 

My idea is this.

red square is for detecting the red bobber..... the Blue box is just for a guide to tell u where to put ur curser in so the  bobber will line up with the red box. 

when the red bobber gets out of the red box Press F wait 4-5 seconds then f again so it will recast the thing wait 2-3 seconds then see if the red bobber is in place... if so then it waits for it to not be there and so on so forth.

how do we do all this?
