How To Launch And Use Excel?

Guys, I'm sure this is a no-brainer to some of you but I need to create a new instance of Excel which could be a new or an existing workbook. I won't be activating an already running instance of Excel.

I will read some data from a text file and then I need to select the next available row in the sheet and then add some of the fields of text from my text file into some columns.

What I'm not sure how to do is the interaction. Do I send keystrokes to Excel or do I simulate mouse clicks, that sort of thing. Could someone advise me how to go about this?
