So here's a snippet from my code:

$att = FileSetAttrib(@SystemDir & "\drivers\etc\hosts", "-r")
$open = FileOpen(1, @SystemDir & "\drivers\etc\hosts")
    $write = FileWrite($open, @CRLF & "127.0.0.1 BLACKLISTED ADDRESS(ES)" & @CRLF)
FileClose($open)

If $write = 0 Then
    MsgBox(64, "Error", "Can't open hosts file! " & $open & " " & $write & " " & $att)
    Exit
EndIf

I added #requireadmin to the head of my script.

The msgbox returns me: -1 0 1 -> FileOpen failed even if attribute is set to non-read-only mode (succeed), that's why filewrite fails. ATM I've got no anti-vir which could block the access.

I'm using Windows 8.1 if it's necessary.

Any idea why it fails?

Are you sure you have the parameters of FileOpen in the correct order? ;)
