Can I use PixelSearch if I do not open the pic

And the all pic files can be coded as X:\*.jpg ,can I?

No you can't

You have to work with GDI32 and GDIPlus Functions

PseudoCode:

GDIPlus -> Load Image
GDIPlus -> Image Get DC
For $x = 0 To Imagewidth
   For $y = 0 To Imageheight
      GDI32 -> GETPixel
      If ColorFound  = ColorSearch Then 
         $foud = 1
         ExitLoop 2
      EndIf
   Next
Next
GDI32 / WinAPI -> deleteDC
GDIPlus -> Dispose Image
if $Found Then Return Array x/y
